yes, this one is very often heard in colloquial

similar construction also:

抽一口烟 / 烟斗 / 烟草 have a smoke of cigarette/pipe/tobacco

吃一口广东点心 have a taste of Guangdong dimsum

other uses of 口:

classifier:

一家五口人 family members (no. of people/population, number of mouths to feed)

三口猪/驴 three pigs/donkeys

一口钢刀 steel sword

一口井 a well (opening)

一口缸 a jar

noun (with or without 儿)

衣服撕了一个口儿 a hole was torn in the jacket

胡同口儿 entrance to hutong

瓶子口儿 mouth of a bottle

碗口儿 rim of a bowl

伤口儿 (opening of) a wound

"他喝了三口水" - "He took 3 gulps of water"?
I don't know if gulp is the most appropriate word here though. I think mouthfuls is probably better as gulp tends to imply a big mouthful. 他喝了三大口水 would maybe be more indicative of a gulp.
